Structural biology seeks to determine the structure of proteins and other macromolecular assemblies in order to understand of their function. Ground-breaking advances in detectors’ technology and image processing is revolutionizing this field by enabling the application of electron microscopy for 3D structure determination. In this talk I will present the principles of data acquisition and image processing for cryo-electron microscopy. The talk will focus on the unique properties of the images produced by the microscope and on the image processing methods which are currently applied in order to reconstruct 3D structures from these images. Emphasis will be given on describing current limitations and future possibilities.
